spectrophotometer analysis of enzymatic solutions
This experiment involved the use of a spectrophotometer and solution mixed by us in the laboratory. The spectrophotometer was used to analyze the speed of the reaction taking place in the sample through the use of light. A solution called guiacol was used because its reactivity with oxygen causes it to turn a brown color adding pigmentation to the solution. (Jacklett 2000) All the solutions were made using guiacol, turnip extract, H202 and dH20. The graphs show how the varying amounts of substrate and enzyme concentration affected the percentage of light absorbed by the solution in the spectrophotometer. This experiment was conducted in order to ascertain the effect of enzyme and substrate concentration on enzymatic reactions taking place in solutions we prepared. An enzyme is a polypeptide macromolecule that catalyzes biochemical reactions ( Bugg 1999). A spectrophotometer was used in order to detect the rate of light absorption by the solution. For the first part of the last sequence of experiments, base line data was used. Part B required us to use 2 times the amount of the substrate. Part C required the use of half the substrate, part D required ¼, part E required 4 times and part F involved 1/10 the amount of the substrate. The other solutions in the tube were changed in accordance with the substrate. The top part of the chart above gives the data to the effects of varying enzyme concentration (Figure #1). The second bottom half of the chart shows the data for the effects of varying substrate concentration (Figure #2).
